Nivalenol (NIV) Testing in Food and Feed

Chromatography-02

What Is Nivalenol (NIV)?

Nivalenol (NIV) is a Type B trichothecene mycotoxin produced primarily by Fusarium species, including Fusarium graminearum and Fusarium culmorum. These fungi infect cereal crops in the field, particularly under cool and wet conditions.
NIV is structurally related to deoxynivalenol (DON) and often occurs alongside DON and other Fusarium toxins in contaminated cereals.
It is most commonly associated with wheat, maize, barley and oats and may be present in both food products and animal feed manufactured from contaminated grain.

Occurrence in Food and Feed

NIV contamination develops primarily during crop growth in the field. Climatic conditions such as prolonged rainfall and moderate temperatures during flowering significantly increase the risk of Fusarium infection and toxin production.
High-risk commodities include:
‣ Wheat and wheat-based products
‣ Maize and maize-based products
‣ Barley
‣ Oats
‣ Cereal-based feed ingredients
‣ Pet food formulations containing cereal fractions
NIV frequently co-occurs with DON and ZEA, particularly in cereal-based raw materials. Because co-contamination is common, single-toxin testing may underestimate overall exposure.

Effects on Animal and Human Health

Like other trichothecenes, NIV inhibits protein synthesis and can affect rapidly dividing cells.
In animals, exposure may result in:
‣ Reduced feed intake
‣ Gastrointestinal irritation
‣ Impaired immune response
‣ Reduced growth performance
Although NIV is generally less studied than DON, it is considered toxic and contributes to the overall burden of trichothecene exposure.
In humans, chronic dietary exposure is controlled through monitoring and risk assessment due to its toxicological profile and potential long-term effects.

The Importance of NIV Testing

Because NIV commonly co-occurs with DON and other Fusarium toxins, comprehensive multi-mycotoxin analysis provides a more accurate assessment of contamination risk.
Analytical testing supports:
‣ Broader assessment of trichothecene exposure
‣ Risk-based raw material evaluation
‣ Protection of livestock performance
‣ Monitoring of cereal-based foods and feeds
‣ Management of co-contamination patterns
Validated LC-MS/MS methods enable simultaneous detection of NIV alongside DON, ZEA, T-2 and HT-2 toxins within a single analytical run.
